Anagrelide, Agrylin Anagrelide causes a dosage-dependant reduction in platelets from decreasing megakaryocyte hypermaturation.  It also inhibits cyclic nucleotide phosphodiesterase and the release of arachidonic acid from phospholipase, possibly by inhibiting phospholipase A2.

Essential thrombocythemia

Before starting Anagrelide, Agrylin make sure your physician is aware of any allergies or medications you currently take, if you have hepatic impairment, are pregnant, or breastfeeding. Anagrelide, Agrylin use has been associated with rare but serious adverse events such as increased risk of major hemorrhagic event, cardiac conductivity disturbances, pulmonary hypertension, pulmonary toxicity, and renal abnormalities.

Oral: Initial: 0.5 mg 4 times daily or 1 mg twice daily and adjust to a platelet count <600,000/mm3 (ideally 150,000/mm3 – 400,000/mm3; usual dose ranges of 1.5 to 3 mg per day).
Maximum single dose: 2.5 mg; maximum daily dose: 10 mg

How to Take: Follow instructions provided by physician/pharmacy label, take consistently at same times each day unless otherwise directed, store at room temperature. May be administered without regard to food.

You Need to Avoid:  Significant drug interactions exist, requiring dose/frequency adjustment or avoidance so avoid starting any new medications without consulting prescriber.  The dose must not be increased by >0.5 mg per day in any one week.

Edema, palpitations, abdominal pain, diarrhea, nausea, dizziness, headache, pain, asthenia
